上海2024年9月26日 /美通社/ -- 北京時間2024年9月27日，愛科百發宣布齊瑞索韋治療嬰幼兒呼吸道合胞病毒（RSV）感染患者的3期臨床研究結果全文發表於《新英格蘭醫學雜志》（The New England Journal of Medicine, NEJM）。這是《新英格蘭醫學雜志》發表的首個中國自主研發的兒童創新藥臨床研究，也是全球首個治療RSV病毒感染靶向藥的3期臨床試驗結果。此次發表由首都醫科大學附屬北京兒童醫院趙順英教授為文章第一作者，首都醫科大學附屬北京兒童醫院倪鑫教授，四川大學華西醫學院附屬第二醫院劉瀚旻教授和愛科百發鄔征博士為共同通訊作者。為此，《新英格蘭醫學雜志》專門發表Edward Walsh教授的社論文章「逐步邁向治療RSV感染的有效抗病毒藥物」，高度評價齊瑞索韋治療嬰幼兒RSV感染的3期臨床研究「結果令人鼓舞」。 RSV是嬰幼兒呼吸道感染住院的首要檢出病原。尤其在冬春季流行季節，RSV感染可導致嬰幼兒發生毛細支氣管炎、肺炎等嚴重呼吸系統疾病，造成高昂的醫療花費，對於早產及患先天性疾病的嬰幼兒甚至可能危及生命，愈後嬰幼兒病人再發喘息，哮喘等呼吸道疾病較健康兒童顯著增加。目前全球范圍內尚無治療RSV感染的特效藥物獲批上市。這項研究積極結果的公布，標志著中國科學家在RSV感染領域取得了「從無到有」的突破性創新成果，走在全球抗RSV新藥研發的前列。這項研究是由首都醫科大學附屬北京兒童醫院倪鑫/趙順英教授團隊牽頭開展，全國30家兒童醫院或綜合醫院兒科參與的臨床3期研究。研究結果顯示齊瑞索韋安全有效，能夠顯著降低RSV感染嬰幼兒的病毒載量，改善臨床症狀。 這項全國多中心、隨機、雙盲、安慰劑對照的3期臨床試驗共納入了311名1~24月齡因RSV感染住院的嬰幼兒患者，以2:1的比例隨機分配到齊瑞索韋治療組和安慰劑對照組，開展為期5天的治療。主要評估指標是治療48小時後，毛細支氣管炎臨床評分較基線的變化。關鍵次要終點是治療96小時後RSV病毒載量較基線的改變。 研究結果： 療效顯著：與安慰劑相比，齊瑞索韋治療組在第一次給藥48小時後的毛細支氣管炎臨床評分平均降低3.4分，而安慰劑組降低2.7分，兩者差異達到0.8分，具有統計學意義（P=0.002）。 病毒載量顯著降低：齊瑞索韋治療組在第一次給藥96小時後的RSV病毒載量較基線平均下降2.5 log10 copies/mL，安慰劑組則下降1.9 log10 copies/mL，差異為0.6 log10 copies/mL，即多降77%（P=0.006）。 安全性良好：齊瑞索韋治療組的藥物相關不良事件發生率為16%，安慰劑組為13%，未發現嚴重的藥物相關不良事件。 研究結果表明，齊瑞索韋作為一款新型口服抗病毒藥物，在治療RSV感染的嬰幼兒中顯示出了積極的臨床效果。藥效主要表現在能夠顯著降低毛細支氣管炎的各項臨床評分，並且降低病毒載量。此外，該藥物的安全性得到了驗證，藥物相關的不良事件發生率與安慰劑組相比沒有顯著差異。 首都醫科大學附屬北京兒童醫院院長倪鑫教授表示：「RSV感染一直是兒科醫療領域的一項嚴峻挑戰，這種病毒不僅對兒童的健康構成威脅，也給醫療系統帶來了沉重的負擔。長久以來，我們一直在尋找一種安全有效的治療手段。今天，我們團隊在這一領域取得了重大進展。我們感謝新英格蘭醫學雜志對這項臨床研究的認可和鼓勵。這是全球首個在RSV感染住院嬰幼兒中開展並取得積極結果的口服靶向抗病毒藥物的3期臨床試驗。我相信，這項研究成果將為全球數百萬受RSV病毒影響的家庭帶來福音，也期待齊瑞索韋早日獲批上市應用於臨床，使中國和全球的RSV兒童患者可以健康成長。」 愛科百發CEO鄔征博士表示：「齊瑞索韋的3期臨床研究結果令人鼓舞，研究證實了齊瑞索韋是一種安全有效的RSV抗病毒治療藥物。作為研究的申辦方，我們深深地感謝所有積極參與這項研究的患者和家長，研究者專家團隊，及各方合作伙伴。我們會繼續努力，與兒童呼吸界專家合作，在國家藥品監督管理局的指導下，推動該藥物的進一步研發和臨床應用。我們將努力不懈繼續致力於兒童創新藥的研發，為兒童患者創造一個更加健康幸福的未來。」 關於齊瑞索韋 齊瑞索韋是一款全新的靶向RSV融合蛋白小分子抑制劑，它通過與病毒的F蛋白結合從而阻止病毒侵入人體細胞。它也可以阻斷病毒通過細胞與細胞間的融合，即形成「合胞體」，一種RSV感染細胞的特征，從而實現抗病毒效果。 齊瑞索韋是全球首個成功完成3期臨床試驗並獲得積極結果的靶向RSV的特效抗病毒藥物，也是首個在中國發明和開發，並拓展到全球的兒童創新藥。同時，齊瑞索韋是首個獲國家藥品監督管理局（NMPA）「突破性治療品種」認定的非腫瘤創新藥。 關於愛科百發 愛科百發是一家創新生物醫藥公司，專注於兒童和呼吸疾病領域未被滿足的重大臨床需求。自2014年成立以來，公司在具有全新作用機制的創新藥物研發領域擁有多項核心技術和全球專利，通過自主研發和外部引進相結合的研發模式，開發出高度差異化的豐富產品管線。核心產品齊瑞索韋 (愛司韋™) 是全球范圍內首款完成3期臨床試驗並取得積極結果的靶向RSV特效抗病毒藥物，也是首個納入國家突破性治療品種的非腫瘤新藥。另一個接近商業化的產品是已獲得FDA批准的兒童多動症新藥AK0901（愛智達™），愛科百發擁有其在大中華區的開發和營銷權。公司還擁有多個具有同類最佳或同類首創潛力的新藥正處於臨床試驗開發階段。 愛科百發與羅氏/基因泰克等跨國藥企、美國學術機構Scripps研究所、中國科學院微生物研究所、國內外生物技術公司，及CRO建立了戰略合作關系。 公司詳情，請訪問網站：www.arkbiosciences.com 投資者垂詢：IR@arkbiosciences.com
